I have the Private Mirror pointing to the env variable DEFS_URL (the virus def. s3 bucket).
When freshclam_update runs, it fails with an 403 error, when trying to download the definitions from the s3 bucket.
Any idea?

Freshclam does not work with IAM permissions. The solution uses a limited scope VPC endpoint with anonymous (no IAM required) access to anything in the scanning VPC to work with this behavior.

You would have to replicate something similar for your private endpoint
